With the introduction of the XLEAP-SBS chemistry, there is a need to select a diverse panel of index sequences (i7 and i5), that will allow for colour balance. The colour balance will help ensure a successful demultiplexing of samples.
This Index Checker is designed to help you determine if the combination you have chosen is ideal for demultiplexing purposes. As a guideline, the proportion of C and T bases combined should take up a minimum of 20% of each position in the index sequence.
How to use the Index Checker
1) Select the number of Singleron indexes and Custom indexes you would like to analyse.
2) Select the Singleron indexes you would like to analyse from the dropdown box.
3) Type in the names of your custom indexes, along with the i7 and i5 sequences. The name and i7 fields are compulsory.
4) Key in the number of reads/GB for each index (please use only either reads or GB, not a mix of both). This will give you a weighted calculation of each index.
5) Click the "Submit" button at the bottom.
6) The table generated will calculate the proportion of C and T bases in each position. If the proportion is less than 0.2 at any position, you are highly recommended to use another combination.
You can use the "Possible UDI combinations" button below to populate a list of Singleron UDI Adapters, where the proportion of C+T is at least 0.2 at each position (assuming equal number of reads assigned).
Any position with "Proportion C+T" less than 0.2 will be highlighted in Red.
It is recommended that your "Proportion C+T" is at least 0.2 at every position before proceeding with this index combination.
